Subcellular distribution of the SVSP454 protein in T. annulata-infected cells. Panels I to V represent the subcellular distribution of the SVSP454 protein in the corresponding cell culture passages (F10, F20, F55, F110, and F165) of T. annulata-infected cells. Panels VI and VII show the positive and negative controls in T. annulata-infected cells, respectively. SVSP454 proteins (panel I to V) and TaSP proteins (panel VI) were stained with rabbit anti-SVSP454 and TaSP polyclonal antibodies, which were each diluted at a ratio 1:100, followed by incubation of the cells with goat anti-rabbit antibody conjugated with Alexa Fluor 488 at a1:1000 dilution. Hoechst 33342 was used at a 1:2000 dilution to label the cell nucleus, and the cytoskeleton was stained with Alexa Fluorâ„¢ 594 phalloidin at a dilution of 1:100.
